該載體通過(guò)自殺基因表達(dá)與否篩選陽(yáng)性重組子。當(dāng)載體與片段連接成功時(shí)，自殺基因無(wú)法正確表達(dá)，包含重組子的細(xì)胞可以正常生長(zhǎng)；當(dāng)載體與片段連接不成功時(shí)，自殺基因正確表達(dá)，包含載體的細(xì)胞無(wú)法生長(zhǎng)，即“Zero”背景。適用于平端克隆。
